Immunochemical studies of crystalline human ceruloplasmin and derivatives.

چکیده
Previous immunochemical investigations of ceruloplasmin have been primarily concerned with the assay of this substance in human sera and its localization by the technique of immunoelectrophoresis. The proteins used in the preparation of antisera appear to have been impure or poorly defined. The ready availability of crystalline human ceruloplasmin (I, 2) prompted us to study its immunochemical properties as an adjunct to its physicochemical characterization (3). The lability of this protein to rather mild treatment is reflected in a pronounced modification of its immunochemical properties. Most of these changes appear to be related to a partial loss of copper from the molecule.

Studies of Human Ceruloplasmin Fractions Separated by Chromatography on Hydroxylapatite.
Recent physicochemical(l,2) and immunochemical(3) studies from our laboratory have shown that human ceruloplasmin is a labile protein the properties of which are readily modified by a variety of experimental conditions. Our previous data, however, indicated that crystalline ceruloplasmin was a single protein. This is at variance with results from other laboratories which have shown that chromat...
متن کاملKinetic studies of ferrous ion oxidation with crystalline human ferroxidase (ceruloplasmin).
The kinetics of the catalysis of Fe(I1) oxidation by crystalline human ferroxidase (ceruloplasmin) has been studied under various conditions at 30’. Two Km values with respect to Fe(U), Km1 = 0.6 /.tM and Km2 = 50 PM, were obtained at pH 6.5, at which ferroxidase gave the maximum activity of 550 Fe(I1) per ferroxidase per min in an acetate buffer. Competition between the two substrates, p-pheny...
متن کاملPros and cons of immunochemical and enzymatic method in the diagnosis of Wilson's disease.
BACKGROUND Immunochemical method of measuring Ceruloplasmin (Cp) levels for the diagnosis of Wilson's disease has replaced enzymatic method for the main reason of being more sensitive and quantitative. SETTINGS AND DESIGN In this study, we compared both the methods for various factors such as sensitivity, specificity and the time consumed in the diagnosis of Wilson's disease. MATERIALS AND ...
متن کاملStudies on the oxidase properties of ceruloplasmin: factors in normal and Wilson's-disease serum affecting oxidase activity.
The oxidase activity of normal serum is due to the presence of the blue copper protein, ceruloplasmin. This is an a-globulin of molecular weight 151,000 containing 8 copper atoms per molecule (1). Its physiological substrate, if any, is unknown (2), but it has a weak oxidase activity for compounds that can be oxidized to quinone or quinone imines (3). The copper atoms in the protein appear to b...
متن کاملA family study of the biochemical defects in Wilson's disease.
Estimations of serum copper, serum ceruloplasmin (immunochemical), and urinary amino-acids excretion (quantitative and chromatographic) in 44 healthy relatives of patients with Wilson's disease (39 from one family) are reported. Each technique revealed some abnormal individuals. Good agreement was obtained between the serum copper and serum ceruloplasmin estimations and between the quantitative...
